The presented method for introducing proteins into living cells should find multifaceted application in cell biology. The pore-forming toxin streptolysin O (SLO) can be used to reversibly permeabilize adherent and nonadherent cells, allowing delivery of molecules with up to 100 kDa mass to the cytosol.

We have approached the development of a vector-/carrier-free method for intracellular delivery of a broad spectrum of cargos by combining exposure to a permeabilizing solution with modified strategies for both applying the permeabilizing solution to the cells and …Author: Shirley O'Dea, Valeria Annibaldi, Louise Gallagher, Joanne Mulholland, Emer L. Dec 09, 2016 · Site-specific fluorescent labeling of proteins inside live mammalian cells has been achieved by employing Streptolysin O, a bacterial toxin which forms temporary pores in the membrane and allows delivery of virtually any fluorescent probes, ranging from labeled IgG’s to small ligands, with high efficiency (>85% of cells).Author: Kai Wen Teng, Yuji Ishitsuka, Pin Ren, Yeoan Youn, Xiang Deng, Pinghua Ge, Andrew S. In one experiment, three different concentrations of a fluorescein-labeled 11-mer (100 nmol/L, 1 μmol/L, and 10 μmol/L) were introduced into reversible permeabilization solutions 2 through 4 to determine whether the distribution of fluorescent ODNs was concentration dependent. This ODN was synthesized by Operon Technologies, Inc.
