A wide variety of absorption enhancers have been evaluated in the delivery of insulin via the ocular route. Some of these are listed in Table 3. As seen in the table, saponin and its derivatives are commonly used as absorption enhancers in the ocular delivery of insulin. The in vivo data indicate that saponin is a strong absorption enhancer.Author: Yung Chi Lee, Pahala Simamora, Sirirat Pinsuwan, Samuel H Yalkowsky

Insulin delivered alone and in the presence of Brij-99 did not significantly alter glucose and insulin concentrations. While effective as an absorption enhancer, 0.5% saponin caused transient ocular irritation in all the test subjects. Brij-99 was also irritating to some of the cats.Author: Rhea V. Morgan, Marcy A. Huntzicker

The ocular route may also be used for the systemic delivery of peptides and protein based pharmaceutical drugs. The ocular route is the choice for the localized delivery of ophthalmologically active peptide and protein for the treatment of ocular disease in the pharmaceutical dosage form of solutions, suspensions and ointment.

    1. Introduction. Peptide and drug delivery through the ocular 1, 2and nasal 3, 4routes of administration has increased in recent years. Of particular interest is the delivery of peptides and proteins through these routes, as it is known that peptidase activity is reduced or minimized in the nasal cavity or the eyes , thus allowing a significant absorption of the bioactive agent.Author: Yoshie Maitani, Tsuneji Nagai, Kyle Kollias, Nikolaos A Peppas
