Jul 06, 2012 · Lentiviruses are a subclass of retroviruses. They have recently been used as gene delivery vectors due to their ability to naturally integrate with nondividing cells, which is the unique feature of lentiviruses as compared with other retroviruses, which can infect only the dividing cells. Lentiviral vectors can deliver 8 kb of sequence.

AAV-based vectors are derived from a normally replication-deficient, non-pathogenic parvovirus and are the smallest viral gene delivery vector capable of transducing both dividing and non-dividing cells. Wild type AAV is a non-pathogenic virus that requires adenovirus or another helper virus for replication.

Viral vectors are tailored to their specific applications, but must generally share a few key properties: Safety: Although viral vectors are occasionally created from pathogenic viruses, they are modified in such a way as to minimize the risk of handling them. This usually involves the deletion of a part of the viral genome critical for viral replication, allowing the virus to efficiently infect cells and deliver the viral …

Non-viral vectors such as cationic lipids and polycationic polymers are used as DNA delivery agents in gene therapy [8,34]. These molecules form electrostatic interactions with DNA, thereby protects DNA from nucleases and cellular defense mechanism.Author: Balamurugan Shanmugaraj, Lohanathan Bharathi Priya, B.
